In english, it can be easily spotted the difference - Day (Singular), Days (Plural), but in Qur'an gives us even more depth.
For example from Surah Al-Fatiha verse 4:
مَـٰلِكِ يَوْمِ ٱلدِّينِ
Maliki yawmid-din
Master of the Day of Judgement
يَوْمِ = Yawm (Day)
Where as, plural example can be seen from Surah Ibrahim verse 5:
وَذَكِّرْهُم
بِأَيَّامِ اللَّهِ
Wa dzakirhum bi ayyamillah
And remind them of the days of Allah
أَيَّامِ = Ayyam (Days)
